The neocortex. Human encephalization is especially pronounced in the neocortex, the most complex portion of the cerebral cortex. The cerebral cortex is the most recently evolved region of the human brain and it is where all high-order cognitive functioning takes place. This is the center of the brain where our self-image and perspective is created.
Only the human brain has evolved to this level; as the human brain evolved, something remarkable happened. The brain, with its great perceptual powers, began to perceive its own existence.
This ability is what makes us human.

The ability to oppose our thumbs at least puts us in the primate category, then the lack of hair on the organ of our skin adds to the illusion that we are human and different from the primates. Then we add the parts of our brain and organ systems that are farther developed.
All in all it is in our DNA, which is in all of our organs and cells which make us most human.
